聚L-谷氨酸酯手性膜制备及其拆分对羟基苯甘氨酸

摘要:
 采用N-羧基内酸酐合成法,分别合成了不同分子量的聚L-谷氨酸甲酯、聚L-谷氨酸乙酯以及聚L-谷氨酸苄酯。将聚L-谷氨酸甲酯、聚L-谷氨酸乙酯以及聚L-谷氨酸苄酯制备成手性固膜,利用扫描电镜表征其结构。研究了不同分子量的聚L-谷氨酸酯、渗析溶剂中不同乙腈含量、原料液浓度、渗析时间、温度等对拆分对羟基苯甘氨酸外消旋体的影响。在优选的实验条件下,这些膜对对羟基苯甘氨酸的手性拆分的e.e.值可达45%以上,且聚L-谷氨酸甲酯>聚L-谷氨酸乙酯>聚L-谷氨酸苄酯。该研究为对羟基苯甘氨酸外消旋体的分离制备,提供了新的研究途径。
 By N-carboxy internal anhydride synthetic methods, different molecular weights of poly L-glutamic acid methyl ester, poly L-glutamic acid ethyl ester and poly L-glutamic acid benzyl ester were synthesized. The poly L-glutamic acid methyl ester, poly L-glutamic acid ethyl ester and poly L-glutamic acid benzyl ester were prepared into enantioselective membranes, and their structures were characterized by scanning electron microscope. The effects of different molecular weight poly L-glutamic acid ester, different acetonitrile content in dialysis solvent, concentration of feed solution, dialysis time and temperature on the resolution of p-hydroxyphenylglycine racemate were studied. Under optimum experimental conditions, the value of e.e. for chiral resolution of these membranes to p-hydroxyphenylglycine can reach more than 45%, and poly L-glutamic acid methyl ester > poly L-glutamic acid ethyl ester > poly L-glutamic acid benzyl ester. This study provides a new way for the preparative separation of p-hydroxyphenylglycine racemate.
