Compare study on pharmacodynamic results and preliminary investigation of refining mechanisms with Gubi granule by using separation technologies of ceramic membrane, ethanol precipitation and other methods

Abstract:
Aim: To investigate three refinement methods and pharmacodynamic results on potency of Gubi granules and explore the feasibility and separation mechanism of separation technology to Chinese herbal compound purified water extract. Methods: Three different refinement methods-ceramic membrane separation, ethanol precipitation and macroporous resin absorption was used. Three types of refined granule products were divided into high-, medium-, low-dose groups . Mice toe-swollen assay, auricle-swollen assay, chondrocyte assay were implemented in order to compare the pharmacological activities of the 9 groups. Result: Three types of refinement products all exerted inhibitory activities against toe-swollen and auricle-swollen. In general, ceramic membranes was thought to have a good anti-inflammatory effect of high-, medium-, low-dose groups in both models.  A significant difference on the growth of value-added rate of chondrocytes was found from three refined products which ceramic membranes was better than ethanol precipitation and macroporous resin. Conclusion: Membrane technology achieved  the refining effect, relatively intacted integrity of Chinese herbal formula at the same time which was thought one of the reason for anti-inflammatory mechanism of ceramic membrane group product. Thus avoiding the loss of the medicinal ingredient of hondrocytes with promoting added value. The ceramic membrane separation technology as traditional chinese medicine water extract purification process solution,  in terms of compound effective substance screening clusters had unparalleled advantage than other separation techniques.
